Yes, I agree. The best place is the high side of the volume pot and ground. That would be considered speaker audio because it is already deemphasized and you would not set deemphasisze in simpleusb.conf.
Use ac coupled audio on the URI. Set rxboost to 1 if you are not getting enough level or 0 if too much. It is nice to try and adjust it somewhere in the middle of the range but not always possible.
The TX audio would go to the mic input. You may need to use you are resistive divider if the level is too high into the transmitter.

The speaker works against ground in that radio.
I wouldn't use speaker output to drive the node. Bump the volume control and your levels are all wrong. Hit it hard enough and maybe damage the input of the USB device.
Top of the volume control would work nicely.

I'm using a midland radio which has AC coupled audio like a Motorola. I am using speaker audio from my midland. I've read you should not ground AC coupled audio because you'll burn up an audio amp. I noticed there is a mic in AC. I am guessing from reading the URI documents that audio goes here.
What do I do with the other lead of the speaker output?
